Czech omitted characters I manually added (red). Issue may be in the encoding of the Czech language. But when creating genealogy this problem is not only when converting to pdf (poster). Thanks so much for your help.

Thank you for details. As the GEDCOM is set in English (this is the reason why your relative is not getting proper Czech characters). To fix the problem export GEDCOM file from your relative and open it in Editor (you will see there LANG English - change it for LANG Czech and save - then import GEDCOM to his/her site - and remove old GEDCOM. This will fix the problem with Czech characters.
